ABSTRACT

Objective To explore the gene frequency of apolipoprotein A5(APOA5)-1131T>C single nucleotide polymorphisms and its relationship with the fat contents of adult Tibetans in Tibet and Han nationality population in Liaoning province.Methods In 100 Tibetan nationality(the study group) and 100 Han nationality individuals(the control group), the polymorphism of apoa5-1131T>C genotypes was detected by polymerase chain reaction-restricted fragments length polymorphism(PCR-RFLP), and fat contents were got by using bioelectrical impedance measurement.Results Results of fat contents of body, trunk and limbs(left upper limbs, left lower limbs, right upper limbs, right lower limbs and total limbs) were respectively below:① There was significant difference between male TC+CC genotype and TT genotype of Tibetan nationality.There was significant difference between male TT genotype of Tibetan nationality and of Han nationality.② There was significant difference between female TT genetype of Tibetan nationality and of Han nationality except result of the body fat contents.③ There was significant difference between the trunk and total limbs fat contents in male TT genotype, and male TC+CC genotype of Tibetan nationality and of Han nationality, and female TT genotype of Tibetan nationality.Conclusion APOA5 gene-1131T>C affects the body fat contents of Tibetans and Han nationality population, and APOA5 gene-1131T>C probably affects lipopexia in trunk.

ABSTRACT

Objective To explore the relationship between digit ratio and personality among male college students of Han nationality. Methods 162 male college students of Han nationality were surveyed with Eysenck Personality Questionnaire. Measurements were taken from the middle of the basal crease to the tip of the finger by electronic vernier caliper. Personality factors scores were compared between male college students of Han nationality and normal. Finger length and digit ratio of the left hand and the right hand were compared among male college students of Han nationality. Correlation coefficient between digit ratio and personality was calculated.Results The male college students of Han nationality showed significantly lower psychoticism (4.53 ± 2. 55 ) and lie (9.64 ± 3. 55 ) than normal ( 5. 96 ± 2.84,12. 17 ± 3.57; t = - 7. 135, - 9. 085 ) , but higher neuroticism ( 12.67 ±3.94) than normal ( 10.63 ±4.44, t=6.573). The length of index finger(2D) & ring finger(4D) and ently higher than those of the right hand (71.74 ± 4. 01 ,74.85 ± 4.41,0. 8973 ± 0. 0306; P < 0.05 ). There was an association of psychoticism with index finger(2D), middle finger(3D), ring finger(4D) and little finger(5D)among male college students of Han nationality ( r=0. 263,0.254,0.248,0.287,0.235,0.243,0.246,0. 283; P< 0. 01 ). There was an association of lie with index finger( 2D ), middle finger( 3D), the right ring finger( 4D ) & little finger(5D) among male college students of Han nationality ( r= -0. 208, -0. 213, -0. 185, -0. 226,-0.165, -0. 185; P< 0. 05 ). There was an association of neuroticism with the left second to fourth digit ratio ratio is found among male introvert college students of Han nationality. Digit ratio may be correlated with personality among male college students of Han nationality.

ABSTRACT

BACKGROUND: Polymorphism in promoter region can change the expression of genes, which may be associated with susceptivity of diseases. Gene polymorphism of interleukin-6 (IL-6) promoter is associated with nationality and many diseases. Different nationalities often display different characteristics of gene polymorphism.OBJECTIVE: To observe the distribution of polymorphism at sites -597 and -572 in IL-6 promoter region in Tibet Tibetan population and to provide the theoretical data for Tibetan population genetics and background of immunity.DESIGN: Randomized investigation.SETTING: Institute of Anthropology, Jinzhou Medical College.PARTICIPANTS: Totally 108 healthy Tibetan teenagers were selected from Lasa and Naqu region in Tibet autonomous region from October 2003 to July 2004, including 60 males and 48 females, aged from 14-21 years. Inclusive criteria:The parents of the volunteers were healthy Tibetans after body examination. The volunteers knew the fact, agreed to participate into the trail and signed the informed consent.METHODS: 5 mL peripheral vein blood was collected from 108 Tibetan teenagers. DNA from human leucocytes was extracted by salt fractionation. IL-6 promoter including -597 and -572 fragments was amplified by polymerase chain reaction (PCR). Representative fragments were cloned then sequenced after restriction fragment length polymorphism (RFLP).MAIN OUTCOME MEASURES: Distribution of polymorphism in Tibet Tibetan population; Results after comparison with those of other nationalities including Han population.RESULTS: Data of 108 Tibetan teenagers were involved in the result analysis. ①Distribution of polymorphism on -572C/G site of IL-6 promoters in population of either sex: There were no GA and AA genotypes at site -597, but only GG genotype appeared. There were CC, CG and GG genotypes at site -572, and the frequencies were 0.63, 0.35 and 0.02 in order. Distribution of genotype with representativeness met the Hardy-Weinberg equilibrium. Allele frequencies were 0.81 and 0.19, respectively. There was no significant difference of either sex in genotype and allele frequency (P > 0.05). ②Distribution of polymorphism on -597 G/A and -572 C/G in different nationalities: GG, GA, AA genotypes appeared on -597 site in England and France, and G and A allele frequencies were 0.60 and 0.40,respectively. It was significantly different from that of Tibetan in Tibet. Furthermore, Japanese had no polymorphism,which was similar to that of Hans in China (P > 0.05). ③Genotype of different straps and results of DNA sequencing:Only GG genotype was found on -597 site (without the restriction site, one fragment after restriction, PCR amplification products), no GA and AA genotypes. CC, CG and GG genotypes appeared at site -572, and frequencies were 0.64,0.35 and 0.01, respectively. Distribution of genotype with representativeness met the Hardy-Weinberg equilibrium (P >0.05). Allele frequencies were 0.81 and 0.19, respectively. There was no significant difference of either sex in genotype and allele frequency. Distribution of gene frequency and allele frequency in IL-6 were similar between Tibetan and that of Hans, but it was significantly different from that of population in England, France and America.CONCLUSION: There are nationality differences of IL-6 gene polymorphism at sites -597 and -572. No polymorphism is found at site -597 in Tibetan. Race differences are seen at site -572, having CC, CG and GG genotypes and G allele is rate. Compared with white population, there is significant difference in genotype and allele frequency at site -572. Their characteristics are close to Hah population and Japanese, which may be associated with genetic gene of persons living in plateau.

ABSTRACT

Objective To investigate the effect of transplanted bone marrow stromal cells on the cells in the subventricular zone of the rats with permanent focal cerebral ischemia and analyze cell types.Methods Permanent focal cerebral ischemia models were established with middle cerebral artery occlusion(MCAO) and divided into three groups: MCAO alone,intravenous infusion of 1ml PBS at 24 hours after MCAO,and intravenous infusion of 2?106 BMSCs 24 hours after MCAO.Then,the groups were subdivided into 7-day and 14-day groups after MACO.Neurological functions were detected by Zausinger evaluation;meanwhile,5-bromodeoxyuridine was injected to label the proliferating cells in the subventricular zone,and double-immunofluscent technologies were used to identify the cell type.Results On the 7th day and the 14th day after MACO,neurological functional scores of BMSCs-treated group were higher than those of the other two groups(P

ABSTRACT

AIM:To evaluate the effect of basic fibroblast growth factor(bFGF)on the expression of cyclin D1,growth arrest,DNA damage inducible gene 153(GADD153),and its roles involved in cell cycle regulation and DNA repair in starvation-induced ovarian cancer CAOV3 cells apoptosis.METHODS:Apoptosis of ovarian cancer CAOV3 cells was induced by serum-free culture(starvation).After bFGF treatment,the cell proliferation rate,cell cycle and apoptosis were determined by MTT,FACS analysis and agarose electrophoresis,respectively.The expression of c-Fos,c-Jun and cyclin D1,GADD153 were detected by Western blotting.RESULTS:bFGF increased the cell proliferation and prevented starvation-induced cell apoptosis.In a time-dependent manner,bFGF induced the expression of c-Fos,c-Jun and cyclin D1 and inhibited GADD153.CONCLUSION:bFGF plays a critical role in anti-apoptosis and the proliferation in human ovarian cancer by upregulating the expression of c-Fos,c-Jun and cyclin D1 and inhibiting GADD153.

ABSTRACT

AIM:To evaluate the effect of bFGF on the apoptosis and the expression of glucose regulated protein 78(GRP78) in ovarian cancer CAOV3 cells.METHODS:Starvation induced cell apoptosis was conducted.After treatment with bFGF,the cell cycle and apoptosis were determined by FACS analysis and Annexin V/PI staining,respectively.The expression of protein kinase B(PKB) and GRP78 were detected by Western blotting and RT-PCR.RESULTS:Compared to starvation group,the cells treated with bFGF were still viable and increased activation of PKB and high expression of GRP78 were observed,which were prevented by PKB inhibitor wortmannin effectively(P
